The principal focus of the Neumann lab at St. Jude Children's Research Hospital is developing novel radiomedicals with diagnostic and/or therapeutic potential toward improving

disease outcomes. We often develop radiomedicals with the goal of translating them into imaging agents for Positron Emission Tomography (PET) to better delineate disease and biological status.

Our lab utilizes synthetic organic chemistry and radiochemistry to synthesize these novel molecules and imaging agents. We then characterize their associated biological activity in relevant cell culture and live animal imaging studies with the goal of clinical translation. We frequently use PET/CT and PET/MRI to evaluate our novel imaging agents in live animals. However, we are now also utilizing Focused Ultrasound techniques to improve therapeutic delivery to the Central Nervous System by reversible

blood brain barrier disruption and sonodynamic therapy. Postdoctoral fellows at St.

Job Responsibilities: Tracking the inventory of chemical compounds, maintaining accurate records, and ensuring the availability of compounds when needed. Managing the storage of compounds in St. Jude's internal repository, which may include researching and maintaining appropriate storage conditions (e. g.

temperature, humidity) and retrieving samples for CBT collaborators as requested. Preparing compounds for distribution to researchers, including dilution, aliquoting, or other sample handling procedures.

Reformatting new compound libraries, purchased or synthesized in-house. This includes the generation of assay-ready plates. Distributing samples to researchers while keeping track of the quantities sent and maintaining proper documentation. Recording and managing data related to compounds, including compound information, storage conditions, and usage records. Having a basic knowledge of analytical techniques used to characterize compounds and verify their identity and purity. Maintaining

up-to-date knowledge of industry techniques, hardware, labware, and software.

Utilizing compound management software to maintain an organized and searchable database of compounds in collaboration with the CBT Lead Discovery Informatics Center, including the internal request system in Share Point. Collaborating with scientists and researchers to understand their specific compound needs and developing protocols to ensure they receive the appropriate samples. Performing experiments to ensure that compounds are stored, handled, and distributed in compliance with safety regulations and best practices. Addressing issues related to compound degradation, contamination, or any discrepancies in compound records by performing experiments.

Collaborating with the CBT Compound Collection team, contributing to designing and organizing compound libraries for high-throughput screening or other applications. Using automation and robotic systems to improve compound management efficiency and accuracy. Reformat new libraries. Training lab personnel on compound management inventory, formatting, and delivery. Minimum Education: Bachelor's degree in relevant scientific area is required. Minimum Experience: Two (2) years of relevant, post-degree work experience is required with a Bachelor's degree.

The Center for Modeling Pediatric Diseases (CMPD) plays a key role in providing that support. As a new initiative at St Jude Children's Research Hospital (SJCRH), its mission is to serve the research community by delivering high quality, fully characterized, patient-derived,

pluripotent cells. Our aim is to generate these cells on our fully automated, state of the art, robotic system. Our methodologies are based on the Nobel Prize winning work of Shinya Yamanaka, which culminated in the ability to create induced pluripotent stem cells (i PSCs) from terminally differentiated human somatic cells.

These i PSCs permit the derivation of both normal and disease phenotype, patient-matched cell lines, and are therefore key to developing personalized medicine approaches. i PSCs provide a vital tool to study the cellular and molecular mechanisms involved in patient-specific responses to treatments, and in the future will form the basis of creating patient-derived,

patient-matched cellular therapeutics. Since i PSCs are derived from somatic cells, they can be created without harvesting vital stem cell pools in the body.

For example, CMPD is able to generate i PSCs from blood and urine, which are collected as part of routine clinical monitoring. We therefore circumvent the ethical issues encountered by working with stem cells from donated oocytes, or embryonic stem cells. As CMPD establishes our methodologies and implements our automation we seek an exceptional and motivated individual who will be able to execute standard operating procedures accurately and efficiently. The transition from manual to automated requires a highly organized, methodical, and algorithmic mindset.

This role provides an exceptional opportunity to learn both manual and automation programming routines. Ability to demonstrate attention to detail is key, as is a desire to work in a highly reproducible manner, by close adherence to optimized SOPs. However, an ability to innovate, and determine where evolutions can be made to improve time and cost efficiencies, in coordination with the team, is also highly desirable. Daily implementation of advanced cell culture techniques, including differentiation protocols in the future, and the ability to undertake sophisticated characterization assays is required.

There are opportunities to learn bioinformatics and data analysis skills, including running machine learning pipelines to evaluate microscope images of developing i PSC colonies. Our automation system includes the latest model instruments: Hamilton Vantage; Hamilton Lab Elite; Cytation 5; Cytomat (500 capacity); King Fisher Flex; Bio Store III. Job Responsibilities The successful candidate will assist the CMPD Director to develop: Daily implementation of advanced cell culture techniques, including reprogramming / biobanking protocols, and the ability to undertake sophisticated characterization assays. Follows SOPs necessary to automate various manual stem cell workflows such as splitting of stem cells, the purification of specific populations, freezing cells, setting up and analyzing various quality control and characterization assays. Assists with the cryopreservation of bar-coded samples using an automated liquid N2 system.

Manages specific, but limited, laboratory functions such as inventory management, ordering, instrument maintenance etc. Demonstrates effort to increase knowledge in relevant areas of research and participates in the presentation and publication of research results, if necessary. Conducts scientific projects and maintains efficient laboratory operations with minimal supervision. Data entry into various databases Assists with stem cell training. Performs other duties as assigned or directed in order to meet the goals and objectives of the CMPD Core and of St Jude Children's Research Hospital.
