The Center for Modeling Pediatric Diseases (CMPD) plays a key role in providing that support. As a new initiative at St Jude Children's Research Hospital (SJCRH), its mission is to serve the research community by delivering high quality, fully characterized, patient-derived,
pluripotent cells. Our aim is to generate these cells on our fully automated, state of the art, robotic system. Our methodologies are based on the Nobel Prize winning work of Shinya Yamanaka, which culminated in the ability to create induced pluripotent stem cells (i PSCs) from terminally differentiated human somatic cells.
These i PSCs permit the derivation of both normal and disease phenotype, patient-matched cell lines, and are therefore key to developing personalized medicine approaches. i PSCs provide a vital tool to study the cellular and molecular mechanisms involved in patient-specific responses to treatments, and in the future will form the basis of creating patient-derived,
patient-matched cellular therapeutics. Since i PSCs are derived from somatic cells, they can be created without harvesting vital stem cell pools in the body.
For example, CMPD is able to generate i PSCs from blood and urine, which are collected as part of routine clinical monitoring. We therefore circumvent the ethical issues encountered by working with stem cells from donated oocytes, or embryonic stem cells. As CMPD establishes our methodologies and implements our automation we seek an exceptional and motivated individual who will be able to execute standard operating procedures accurately and efficiently. The transition from manual to automated requires a highly organized, methodical, and algorithmic mindset.
This role provides an exceptional opportunity to learn both manual and automation programming routines. Ability to demonstrate attention to detail is key, as is a desire to work in a highly reproducible manner, by close adherence to optimized SOPs. However, an ability to innovate, and determine where evolutions can be made to improve time and cost efficiencies, in coordination with the team, is also highly desirable. Dr. Tsai is co-leading a new Blue Sky Project called PARADIGM (Partnership to Advance Development of Individualized Genomic Medicines) with goal of developing personalized genome editing therapies for St. Jude patients. You would have opportunities to lead exciting projects that include: Protein engineering of improved novel CRISPR base and prime genome editors with human cell-based directed evolution. Understanding the structure-function relationship of mutations identified from high-throughput screens on the
activity of genome editors. Perform deep mutational scanning of CRISPR-Cas orthologues and effector domains important for precise genome editing such as deaminases and reverse transcriptases.
Define the outcome of engineered variants on genome editing repair outcomes using high-throughput genomic methods. Help develop laboratory automation workflows to perform scalable protein engineering. You would learn valuable laboratory skills that include: Genome engineering, high-throughput genomics, next-generation sequencing, protein engineering, structural biology, liquid handling automation, hematopoietic stem cell cell culture and transfection, and data science.
Project-dependent responsibilities could include: Engineer CRISPR base and prime editors with improved activity and specificities. Test new strategies to edit human hematopoietic stem cells for treatment of bone marrow failure disorders, focused on rapid development of editing strategies for rare patient-specific mutations. Optimize lead protein candidates by structure-guided design and high-throughput screening. Apply new machine learning techniques to guide protein engineering design. Analyze experimental outcomes and develop high-quality summaries towards manuscripts and grant applications.
